MYB106 is a negative regulator and a substrate for CRL3 <sup>BPM</sup> E3 ligase in regulating flowering time in <i>Arabidopsis thaliana</i>

Flowering time is crucial for successful reproduction in plants, the onset and progression of which are strictly controlled. However, flowering a complex environmentally responsive history trait underlying mechanisms still need to be fully characterized. Post-translational regulation activities transcription factors (TFs) dynamic essential mechanism plant growth development. CRL3BPM E3 ligase CULLIN3-based involved orchestrating protein stability via ubiquitin proteasome pathway. Our study shows that mutation MYB106 induced early phenotype while over-expression delayed Arabidopsis flowering. Transcriptome analysis myb106 mutants reveals 257 differentially expressed genes between wild type myb106-1 mutants, including Locus T (FT) related time. Moreover, vitro electrophoretic mobility shift assays (EMSA), vivo chromatin immunoprecipitation quantitative polymerase chain reaction (ChIP-qPCR) dual luciferase demonstrate directly binds promoter FT suppress its expression. Furthermore, we confirm interacts with BPM proteins further identified by ligases as substrate. Taken together, have negative regulator control new substrate Arabidopsis.
